In afforestation programs, both for productive and conservation purposes, the use of the correct seed source is a key factor. The Nothofagus obliqua ‘Trevelin’ seed production area (SPA) was installed during 1997 at INTA Agroforestry Station, Chubut province. ‘Trevelin’ SPA has two different origins. While some individuals were produced with seeds belonging to Lácar forests in Argentina (Hua Hum provenance), the rest of the seedlings were produced with seeds collected from a stand (EAFT provenance) with unknown origin. In the present work, we tried to identify the origin of the individuals at the ‘Trevelin’ SPA, analysing the chloroplast DNA (cpDNA) of the mother trees used for their production (EAFT provenance). Based on the clonal and uniparental inheritance of cpDNA, it represents a useful tool in the definition of large groups of populations with a common gene pool. In most angiosperms, this marker enables the identification of lineages of maternal inheritance. The comparison of the results with previous genetic population information allowed the origin identification. For this purpose, we sequenced two inter-genic regions of the chloroplast DNA, trn-D and trn-T and ath-H and atp-I, in 12 mother trees. The analysed samples showed haplotype I, which corresponds to the southern lineage populations of the species in Argentina, with forests distributed in the Lácar basin. Therefore, it is possible to prescribe the use of the seed of this stand, not only in actions of commercial plantation outside the natural distribution area of the species, but also, in restoration activities of N. obliqua forests within Lácar basin. Even more, direct associations could be established between the haplotypes found by the PCR-RFLP technique (used in previous studies) and the sequencing (used in the present study) for both inter-genic regions of the analysed chloroplast DNA. This will allow using the method of sequencing in future analyses without losing the homology with the information obtained by the oldest method.
