Abstract

Prosopis limensis Benth.in Hook is a native tree species that grows in the seasonally dry forest and is important for firewood and forage production and environmental protection. The results of this research were submitted to variance analysis (ANOVA) of a single factor, nonparametric Kruskal-Wallis and Tukey's test. In vitro seed germination with physical and chemical scarification was 100% whilw rooting of cuttings was of 0%. In tissue cultures, shoot-tip of 5 cm height seedlings obtained from in vitro germinated seeds, reached the highest bud elongation in MS culture media supplemented with 0.2 mg L-1 KIN, showing significant differences only about in relation to 0.2 mg L-1 BAP and 2.0 mg L-1 KIN. In other treatments tested, i.e. 0.2 mg L-1 NAA + 0.2 mg L-1 KIN, 0.2 mg L-1 IAA + 0.2 mg L-1 KIN and 0.02 mg L-1 IAA + 0.02 mg L-1 GA3 although increased bud elongation was reached, the results were not significant.The number of shoots formed was not more than 2, in the culture medium supplemented with 1.0 mg L-1 KIN + 25 mg L-1 adenine sulphate, while rooting was better in the culture medium supplemented with 1.0 mg L-1 IAA. The survival rate during greenhouse acclimatization was of 90%. This paper reports a simple but effective method to micropropagate P. limensis from apical shoots and nodal segments of seedlings obtained by in vitro germinated seeds, since although the sprouting rate was low, the number of nodes formed per seedling was significantly higher.
