Abstract

Cellulose is the main component of the cell wall with great biotechnological importance regarding the analysis of new cellulase producers microorganisms. The objective of this study was the design and testing of a submerged production system of endo-β-1.4-glucanase using pine chips as substrate; pH and T optimum action of the enzyme extracts by two white rot fungi Irpex sp. and Phlebia sp. were checked at laboratory scale. The production system proposed in this study showed a logarithmic behaviour with respect to enzyme production with maximum activity of 1 U / Lh for Irpex sp.strain and 1.37 U / Lh for Phlebia sp. strain. Enzymatic activity remained constant over time through washing and adding fresh medium, simulating a fed-batch production system. Extracts of both strains showed better performance at 50°C, with higher activity at pH 3 for extracts of Irpex sp. strain and pH 4.5 for Phlebia sp. strain. This system may be optimal to start scaling up studies.
