Yellow leaf disease, caused by Sugarcane yellow leaf virus (SCYLV), is worldwide spread in sugarcane regions, however; its incidence and distribution in Argentina it s unclear. The two main aims of this study were to: i) analyse SCYLV distribution in the sugarcane production area in Tucumán, characterize the virus and determine the correlation between virus presence and disease symptoms, and ii) evaluate the behavior of progenitor collection of the Sugarcane Breeding Program (SBP) of the Estación Experimental Agroindustrial Obispo Colombres (EEAOC) against the virus, using different diagnostic techniques. In 2011 and 2012, 146 sugarcane leaf samples with and without yellowing symptoms were collected in Tucumán Province from commercial varieties, advanced breeding clones at final testing stages of selection and progenitors of the SBP. SCYLV was detected by RT-PCR in 26 samples, all of them corresponding to BRAPER genotype. Phylogenetic analysis of the SCYLV-CP gene allows clustering of strains as well as to distinguish among individual isolates. The correlation between virus detection and disease symptoms was positive and low but statistically significant. Regarding the second goal, samples from parents used in SBP were collected during 2013, 2015, 2016 and 2017 and diagnostic techniques, molecular (RT-PCR) and serological (TBIA) were compared. Seventy five percent of the genotypes remained disease-free throughout the experimental period. Respect to the diagnostic methods, it is concluded that RT-PCR using SCYLVf1/r1 primers would be more sensitive than YLS111/462 pair, and that molecular diagnosis was more sensitive than serologic one.
