The Schinopsis balansae is a typical tree of the Oriental Chaco whose growth is relatively slow; its seeds are produced quite unevenly yearly and among individuals so that germination, vigor and health show great variation. The aim of this study was to establish an in vitro seed germination protocol and an in vitro culture of S. balansae out of three month seedlings. Fruits were harvested in Vera, Santa Fe and their seeds extracted and disinfected using alcohol 96% by immersing them into a 2% sodium hypochlorite solution added with 2 drops of Tween 20 for 10 minutes were afterwards sown on water-agar (AA). Seedlings were transferred to the Murashige and Skoog medium: 1-100% (MSE) and 2-50% (MS 0.5). Germination percentage in AA and number of leaves, stem and roots length were evaluated 30 days after being transferred. Explants from the in vitro seedlings were extracted and sown in three MS media at different concentrations of BAP and ANA. Callus regeneration and necrotic explants that exuded tannins were evaluated. It was obtained 69% of normal seedlings with no growth differences among means. The percentage of differentiated callus varied significantly among means, being those in the 2 mg.L-1 BAP and 1 mg.L-1 NAA media the maximal differentiation observed. The number of explants exuding tannins and of necropsied explants varied according to the culture.
