This work is based on the need to have a method for the isolation of lignocellulosic fibrous material for microscopy, without damaging the cells so much, which can be carried out in a short time in the laboratory, at atmospheric pressure, and without toxic. A procedure which consists of treating wood with sodium hydroxide into monoethanolamine at 170o C for 1 to 4 hours was developed. A variety of woods could be isolated- softwoods as well as hardwoods. The amount of alkali used was 25% on dry wood, with a liquor-wood relationship of 10. A quantitative analysis of fibers gave as a result between 4 and 5% of broken elements. A direct relationship between wood density and the time required to complete the treatment was found, being of 1 hour for the 0.14 Kg/dm3 density, and of 4 hours for the 0,80 Kg/dm3 density.
