In vitro mitogen induced lymphocyte proliferation test is widely used for evaluation of cellmediated immunity. In the present work, we compare the results of the traditional method, which uses gradient isolated lymphocytes as the source of cells for the culture and tritiated thymidine incorporation to measure proliferative response (LP- 3H), with other two methods: culture of whole blood and proliferation measure with 3H- thymidine (SE-3H), and culture of isolated lymphocytes and proliferation evaluation with a colorimetric assay using MTT (LP-MTT). Blood samples of twelve adult, healthy dogs were processed by the three mentioned methods, using Concanavalin- A as mitogen and the Stimulation Index (SI) to express the degree of lymphoproliferation. The SI obtained by the SE-3H method were significativily higher than those of the traditional test, while those obtained by the LP-MTT method were significativily lower. These results show that the SE- 3H method could be a suitable alternative to evaluate the lymphoproliferation in our laboratory conditions; it is required more research to evaluate if, in spite of the lower SI, the LP-MTT method could be suitable to evaluate lymphoproliferation in dogs. Based on the results obtained in this experience, we established a normal SI range for each method.
