In Mexico, the state of Oaxaca has occupied the first places in the national level in the production of papaya (Carica papaya L.) in consecutive years. On the other hand, plantations are highly susceptible to viral attack, one of the most devastating is the papaya ringspot virus (PRSV). For that reason, three specific oligonucleotides were designed that allow the identification of the PRSV using the reverse transcriptase coupled polymerase chain reaction (RT-PCR). The oligonucleotides were designed by Vector NTI Advance ® V11.5 software that amplified three fragments of 857 base pairs (bp), 583 bp and 213 bp from conserved regions of the viral genome. To check the performance of the oligonuceotides, ribonucleic acid (RNA) extractions of papaya leaf tissue with symptoms of PRSV were performed using TRIzolTM, then the SuperScriptTM III One-Step RT-PCR System Kit was used to perform the RT-PCR technique. The results of the amplification were analyzed on an agarose gel where the amplicons coincided with the size expected at their preliminary design.
