White rot fungi, such as Trametes villosa BAFC 2755, are the only organisms capable of efficiently degrading lignin due to their oxidative enzymatic system. One of the most important enzymes in this process is lacease. This is a phenol oxidase, which has applications in several biotechnological processes including pulp delignification, detoxification of recalcitrant chemicals, degradation of polycyclic aromatic compounds, bioremediation of contaminated soils and waters. The objective of this work was to evalúate the effect of pH, temperature and incubation time on the growth and oxidative potential of T. villosa BAFC 2755, for that were carried out a quantitative of tests in culture médium ME and activity in lacasa were determinated, to complete the analysis a gene fragment of this enzyme were characterize. The results obtained showed that the conditions that improved biomass production were not those that supported the greatest activity lacease. Maximun activity levéis (200 U/L), were recorded after 14 days, at pH 5.5 and 25° C. Lacase's isoenzyme profile was affected by pH, temperature and culture time, with three isoenzymes of 64 kDa, 123 kDa and 132 kDa. In addition, a nucleotide sequence of 779 bp was obtained from a gene coding for lacase, whose protein sequence showed 90% identity with sequences belonging to the lacase gene of other species of Trametes sp.
